Project Details: In this project, we will generate novel transgenic mouse models to study mechanisms of platelet receptor (GPVI, CLEC-2) signalling in the settings of thrombosis, haemostasis and thrombo-inflammatory diseases. A CRISPR/Cas9 based gene editing platform will be established (in Würzburg) to test basic mechanisms of early ITAM receptor signalling steps identified within the TAPAS consortium; and to validate targeting strategies to modulate the function of these receptors in vivo (Würzburg) and in vitro (Maastricht). This ambitious project will combine state-of-the-art gene editing technology and in-depth biochemical and cell biological studies on platelet receptor functions from single cell to whole body. This project will closely interact with other projects and will provide an essential translational platform within the TAPAS consortium.
